Book the dome climb before you arrive
The Duomo has two different lines in one building. The cathedral is free and has a long line to show for it. The dome climb is timed, capped and sells out days ahead in season.
Book the dome climb before you travel and take the first slot. For the free cathedral, the line is shortest at opening and late in the day.
Uffizi and Accademia: first slot, not Monday
The Uffizi and the Accademia are closed on Mondays. At the Uffizi a walk-up ticket can cost two hours in line in season. At the Accademia the reserved line moves; the unreserved one stands in the sun. The street from the Duomo past the Signoria to the Uffizi fills from 10:00 and eases after 17:00.
Reserve both. Take the first slot at the Accademia and be out by 10. At the Uffizi, go at opening or after 16:00 and see Botticelli first or last. Cross the Ponte Vecchio at dawn or after dinner.

Florence's crowd keeps to the street from the Duomo past the Signoria to the Uffizi, the Ponte Vecchio all day, the Accademia's unreserved line and Piazzale Michelangelo at sunset. The Oltrarno, Santo Spirito and San Frediano, the Bargello and San Marco, the upper terraces of the Boboli gardens and San Miniato al Monte stay quiet.
Stay a few stops from the centre, near one of the quiet places. The Oltrarno is calm all day. The city page names the quiet places and links to hotels there.
Pack for first slots and a dawn walk
In September Florence has about 27 °C by day and rain on about 9 days. The tips above ask for first slots, a dawn crossing of the Ponte Vecchio and a lot of walking.
